Within the Matthias Schleiden Institute’s Division of Genetics, led by Prof. Theißen, the Faculty for Biological Sciences is seeking a Doctoral Researcher in Plant Biology. The position starts from March 1, 2024, or later, until it is filled.

Our research group specializes in evolutionary developmental biology of plants, emphasizing MADS-domain transcription factors, miRNAs, as well as flowers, fruits, and seeds. The primary objective of our new project, titled “MADS life on MAdLand II: Changes in DNA-binding specificity of MADS-domain transcription factors during the transition of plants to land,” aims to comprehensively grasp the role of MIKC-type MADS-domain transcription factors (MTFs) in the plant’s transition to land.

Previously, we’ve observed significant alterations in protein complex formation and DNA-binding between MTFs from charophyte algae and land plants. Our ongoing research will delve into understanding their mutational basis, biophysical attributes, gene regulatory impacts, and resulting phenotypic consequences. This project is an active participant in the DFG priority program 2237, “MAdLand – Molecular Adaptation to Land: plant evolution to change.”

  • Study DNA-binding specificity of charophyta, Marchantia, and Physcomitrium MTFs using competitive EMSA, SELEX-seq, and site-directed mutagenesis.
  • Conduct ChIP-seq to identify DNA-binding sites of a mutant MTF in Marchantia polymorpha compared to wild-type proteins.
  • Assist in writing publications and work reports alongside laboratory work.
  • Perform RNA-seq and analyze differential gene expression using wild-type plants, MTF knock-out plants, and CRISPR-Cas9 edited mutants of M. polymorpha.
  • Identify DNA-binding sites of MTFs in charophyte species’ genomes through DAP seq.
  • Reconstruct the evolutionary path of an ancestral MTF’s DNA-binding specificity across land plants.
